A Lithium-sensitive Regulator of Sperm Flagellar Oscillation Is Activated by cAMP-dependent Phosphorylation

چکیده

Specific effects of both in vivo activation and in vitro activation by cAMP-dependent phosphorylation on bending wave parameters of demembranated, reactivated, tunicate (Ciona intestinalis) and sea urchin (Lytechinus pictus) sperm flagella can be reversed by exposure to protein phosphatase. The effects of protein phosphatase incubation can be imitated by inclusion of LiCI in the reactivation solutions. The primary effect of cAMP-dependent phosphorylation appears to be activation of a regulatory mechanism controlling flagellar oscillation, rather than activation of the active sliding mechanism. Lithium appears to act on the same regu-

O-11: Dynamics of Flagellar Force Generated by A Hyperactivated Spermatozoon

Background: To clarify the mechanism of sperm penetration through the zona pellucida, the flagellar force generated by a hyperactivated spermatozoon was evaluated using the resistive force theory applied to the hyperactivated flagellar waves that were obtained from the mammalian spermatozoa. Materials and Methods: The hydrodynamic calculation of the flagellar force of the activated (non-hyperac...

Axokinin phosphorylation by cAMP-dependent protein kinase is sufficient for activation of sperm flagellar motility

Using a selective inhibitor of cAMP-dependent protein kinase, N-[2(methylamino)ethyl]-5-isoquinolinesulfonamide (H-8), the requirement for cAMP-dependent phosphoproteins in the initiation of dog sperm flagellar motility was examined. H-8 inhibited motility of live as well as reactivated sperm in a dose-dependent manner. The half-maximal inhibition of reactivated motility (32 microM) paralleled ...

Distinct Roles of Soluble and Transmembrane Adenylyl Cyclases in the Regulation of Flagellar Motility in Ciona Sperm

Adenylyl cyclase (AC) is a key enzyme that synthesizes cyclic AMP (cAMP) at the onset of the signaling pathway to activate sperm motility. Here, we showed that both transmembrane AC (tmAC) and soluble AC (sAC) are distinctly involved in the regulation of sperm motility in the ascidian Ciona intestinalis. A tmAC inhibitor blocked both cAMP synthesis and the activation of sperm motility induced b...

Increased intracellular pH ([pH]i) activates dynein in sea urchin and mammalian sperm and induces activation of flagellar motility. It is thought that cAMP-dependent protein phosphorylation is associated with motility

Eukaryotic cilia and flagella are found on various types of cells, from single-cell protozoa to epithelial cells of multicellular organs. Most have a well-conserved structure called the axoneme, which generates ciliary and flagellar movements. The axoneme is a microtubule-based, highly organized structure composed of more than 250 kinds of proteins. Molecular motors, called the dyneins, exert f...
